Structural dynamics of H/ACA ribonucleoproteins studied by single molecule Förster resonance energy transfer spectroscopy

H/ACA ribonucleoproteins catalyze the modification of uridine to pseudouridine nucleotides in ribosomal and spliceosomal RNA of Archaea and Eukaryotes. To significantly expand our understanding of their molecular mechanisms, this project will use complexes reconstituted from labeled recombinant proteins and synthetic RNAs, with the option to add isolated native (sub-) complexes. Single molecule FRET spectroscopy will be employed to investigate and compare structural dynamics of archaeal and yeast H/ACA RNP complexes during assembly and catalysis, and opens the possibility to study complexes comprising one (archaeal) or two (yeast) functional units.
